Lipase from Candida cylindracea was immobilized on chitosan beads by c
ovalent binding methods, and its application to hydrolysis of beef tal
low was investigated in a biphasic organic-aqueous system. The additio
n of isooctane, found to be the best among the organic solvents tested
, to the reaction system stimulated the hydrolysis of beef tallow by i
mmobilized lipase strongly. The proportion of isooctane and aqueous bu
ffer in the reaction mixture was chosen at the ratio of 1 : 4. Under t
his condition, the reaction rate was greatly enhanced compared to that
in the organic-solvent-free system, and the extent of hydrolysis reac
hed to about 90%.
