Human placenta 40S ribosomal initiation complexes were obtained using
40S ribosomal subunits and eIF-2 . GTP . Met-tRNA(f)Met and then modif
ied with O-[4-N-(2-chloroethyl)-N-N-methylamino]benzylidene derivative
s of AUGU(n)[P-32]pC and hloroethyl)-N-methylamino]benzylmethylphospho
amide derivatives of [P-32] pAUGU(n) oligoribonucleotides. After the i
nteraction with these mRNA analogs, the modified ribosomal proteins we
re analyzed. It was shown that in the presence of the ternary eIF-2 .
GTP . Met-tRNA(f)Met complex, the S3/S3a, S6, S7, and S14/S15 proteins
play the key role in the 40s subunit-template interactions; the S2, S
4, S5, S8, S9, and S17 proteins are also involved.
