Radioactive PAPS was synthesized with a predetermined specific activit
y using enzymes purified from Bacillus stearothermophilus. The reactio
n system, incorporating 1.5 mCi of S-35-H2SO4 and 500 nmol of ATP, pro
duced 582 muCi of S-35-PAPS. The synthesized S-35-PAPS was purified by
ECTEOLA cellulose chromatography, followed by desalting with DOWEX 50
W-X8. The final yield was 206 mumol of S-35-PAPS with a radioactivity
of 411 muCi.
