6-Aminohexylated macroporous glucomannan gel beads (AH-GM gel) have be
en developed and applied to chromatographic separation of blood coagul
ation factor VIII(FVIII) in plasma cryoprecipitate fraction. Retained
capacity of FVIII on various kinds of AH-spherical gels was measured b
y a batch-wise method and was apparently dependent on exclusion limit
(Mlim) of the gels. AH-GM gel (MliM 1 x 10(7)) possessed the capacity
at least 10 fold higher than EAH Sepharose(R) 4 B. The HPLC system usi
ng an AH-GM gel column also showed strong retention and good elution r
ecovery of FVIII under the operational conditions at 0.5-4 m/h flow ra
te.
