Expression constructs for human endothelin (ET) receptors ET(A) and ET
(B) were made by subcloning the corresponding coding sequences into th
e pMPSV/CMV vector. They were used together with plasmids bearing resi
stances to puromycin and hygromycin for transfection of baby hamster k
idney cells. Culture in the presence of both antibiotics allowed the s
election of cell lines stably expressing one receptor or the other. Co
mpetitive binding experiments using ET-1 and ET-3 showed the typical i
sopeptide-selective and non-isopeptide-selective profiles for recombin
ant ET(A) and ET(B), respectively.
