The ability of alpha-mannosidase and alpha-galactosidase to cleave the
alpha-linkage between mannose or galactose and serine or threonine wa
s studied The following glycopeptides were synthesized as model compou
nds simulating the O-glycosyl linkage in glycoproteins: utoxycarbonyl-
O-alpha-mannopyranosyl-seryl-glycine methylamide (alpha-Man-Ser-Gly),
xycarbonyl-O-alpha-mannopyranosyl-threonyl-glycine methylamide (alpha-
Man-Thr-Gly), nosyl-seryl-glycinemethylamide(alpha-Gal-Ser-Gly), and b
utoxycarbonyl-O-beta-mannopyranosyl-seryl-glycine methylamide (beta-Ma
n-Ser-Gly). The cleavage was shown to occur in glucoamylase after prot
eolytic degradation.
