Glucose oxidase in monomolecular films obtained from Penicillium vital
e based on polymeric matrices were studied with a Clark-type oxygen el
ectrode. The kinetic constants of the immobilized and dissolved forms
of the enzyme were compared. The kinetic activity of glucose oxidase i
n monomolecular films was assayed under various conditions of applicat
ion, measurement and storage. The data obtained indicate that glucose
oxidase immobilized on positively charged surfaces (polyethyleneimine)
allow the construction of a glucose electrode with a linear response
in the range of 1-10 mM and a response time of 20 s. The formation of
monomolecular glucose oxidase films results in a significant decrease
of the Michaelis constant. The glucose electrode is capable of maintai
ning the initial enzymic activity over 2 months.
