Abstract
The RFLP analysis of rice with cDNA and genomic DNA probes was made in two cultivars of rice plants, Oryza sativa L. Aikoku (Japonica) and C ulture 340 (Indica). Among 114 DNA probes obtained from PstI digested genomic DNA of Culture 340, 62 showed RFLPs with at least one of four restriction enzymes, BamHI, EcoRI, PstI and XbaI. RFLP detection level (% of probes which showed polymorphism) was nearly 25% except PstI (8 .7 %). There was strong relationship (r = 0.99*) between RFLP detecti on level and the observed average size of restriction fragments in eac h enzyme. On the other hand, among the 76 cDNA probes made from Japoni ca var. Yamahoushi, 63 showed RFLPs with at least one of four restrict ion enzymes, where RFLP detection level was 51-59 % in all enzymes. Mo reover, there was no relationship between RFLP detection level and the observed average size of restriction fragments in each enzyme. Thus, cDNA probes are capable of producing high RFLP detection level than ge nomic DNA probes.
