Tissue eosinophils can be easily and specifically demonstrated in froz
en sections after substrateless incubation with 3,3'-diaminobenzidine
(DAB) at neutral pH. Under such conditions, other cells possessing end
ogenous peroxidatic activity (neutrophils, macrophages) do not stain,
whereas staining of mitochondria-rich cells due to cytochrome oxidase
activity can be avoided by a short prefixation of sections in glutaral
dehyde-containing fixatives. The method can be useful in cases when a
rapid screening of tissue eosinophils is required.
