M. Patek et al., PRODUCTION OF THREONINE BY BREVIBACTERIUM-FLAVUM CONTAINING THREONINEBIOSYNTHESIS GENES FROM ESCHERICHIA-COLI, Folia microbiologica, 38(5), 1993, pp. 355-359
Genes of the threonine operon of Escherichia coli were used for the co
nstruction of a Brevibacterium flavum strain excreting threonine. Usin
g the shuttle vector pCEM300 and a newly constructed shuttle vector pE
C71 (7.1 kb, Km(r)/Nm(r)), various plasmids carrying E. coli thr genes
were prepared. Mutants resistant to the threonine analog 2-amino-3-hy
droxyvaleric acid (AHV) were isolated after the ethyl methanesulfonate
treatment of B. flavum carrying these recombinant plasmids. A mutant
B. flavum CCM 351 carrying the cloned genes thrA and thrB accumulated
12 g/L of threonine after 48 h of cultivation.
