In order to perform melphalan dosage adjustment, the linearity of melp
halan kinetics was studied, in the case of previous carboplatin admini
stration. Eleven patients with various solid tumors entered the presen
t study. Carboplatin was administered during 5 days over 1-hour infusi
ons; the day after, the melphalan test-dose was administered and follo
wed 24 hours after by the complement dose. Melphalan kinetics were det
ermined from only three plasma samples by using bayesian estimation. T
he present study showed that previous carboplatin administrations indu
ced wide variations of melphalan pharmacokinetic parameters between th
e two administrations. As part of this protocol, the order of drug adm
inistration should be taken into account, in order to perform melphala
n dosage adjustment.
