Abstract
A sandwich enzyme immunoassay using a chemiluminescent detection has b een developed for the quantification of total human (high and low mole cular weight) kininogens in tissue extracts. This assay uses monospeci fic polyclonal IgG labelled with alkaline phosphatase and the commerci ally available dioxetane derivatives as substrates, for the detection of immune complexes. This method exhibits a sensitivity level of 1 fmo l/ml and allows a precise quantification of total kininogens in synovi um and cartilage extracts. When characterized by Western blot, the imm unoreactive material reveals the presence of both high and low molecul ar weight kininogens.
