PCR-AMPLIFICATION OF MIXED 16S RIBOSOMAL-RNA GENES FROM AN ANAEROBIC,CYANIDE-DEGRADING CONSORTIUM

The microorganisms participating in the anaerobic biodegradation of cy anide were characterized using 168 rRNA genes as genetic markers of di versity. Segments of mixed population 16S rRNA genes were amplified us ing the polymerase chain reaction (PCR) and prokaryote-specific amplif ication primers. Restriction fragment length polymorphisms (RFLPs) and screening with the 926f universal sequencing primer were used to cate gorize the cloned PCR products. Six unique prokaryote sequences were o btained, including four similar to methanogens and two similar to Gram -positive eubacteria.
