A lambda ZAPII-cDNA library constructed from poly(A)(+)-RNA isolated f
rom Fe-deficient barley roots was used for differential screening of b
arley roots grown in the presence and absence of Fe. Among seven clone
s that hybridised specifically to the probe for Fe deficiency, one clo
ne (Ids2) was sequenced. Using a part of the cDNA sequence as a probe,
a genomic-DNA library was probed and a corresponding DNA clone was is
olated and sequenced. The predicted amino acid sequence resembled 2-ox
oglutarate-dependent dioxygenase. Ids2 had a metal regulatory element
as well as Cu regulatory elements of CUP1 gene of yeast MT.
