A physical chromosomal map of Acholeplasma oculi ISMI-499 was construc
ted by using field inversion gel electrophoresis. To assist in the ord
ering of the chromosomal fragments, a modified transposon, Tn4001.1064
, was constructed. It was also used to rescue mycoplasmal chromosomal
sequences adjacent to transposon insertion sites in a one-step cloning
procedure. The total size of the A. oculi ISM1499 genome was estimate
d to be 1,633 kb. The restriction enzyme sites for ApaI, BssHII, EagI,
and SmaI were positioned on the map along with several transposon ins
ertion sites.
