Abstract
We describe a method for anchoring bacteriophage lambda DNA by one end to gold by Au - biotin - streptavidin biotin - DNA bonds. DNA anchore d to a microfabricated Au line could be aligned and stretched in flow and electric fields. The anchor was shown to resist a force of at leas t 11 pN, a linkage strong enough to allow DNA molecules of chromosome size to be stretched and aligned.
