A novel procedure for isolation of acid-resistant urine trypsin inhibi
tor (UTI) is described The method is based on the use of UTI-specific
monoclonal antibodies immobilized on an insoluble matrix. Utilization
of Immotin (UTI immobilized on Sepharose 4B) as a precolumn allows con
siderable increase in the sen,ice-life of the immune sorbent. Isolatio
n of UTI by this procedure increases the yield up to 92% of original u
rinary content of the inhibitor. The resulting preparation contains pr
edominantly the native form of the inhibitor (-44 kD molecular weight)
