A new method for measuring the freezing denaturation of ovalbumin with
5,5'-dithiobis-(2-nit robenzoic acid) (DTNB) is described. One ml of
an ovalbumin solution (0.1%) containing 50 mu M DTNB at pH 7.0 was sub
jected to 5 freeze-thaw ((FT) cycles. After centrifugation of the solu
tion, the absorption at 412 nm was measured as an index of denaturatio
n. This method was applied to study the cryoprotective effect of sucro
se. The method offers a simple and accurate means for measuring the de
naturation of ovalbumin produced by FT cycles and for measuring the ef
fect of cryoprotectants on this denaturation.
