NotI linking clones represent valuable tools for both physical and gen
etic mapping. Using procedures that we have previously described, seve
ral chromosome 3-specific NotI linking libraries have been constructed
. Here, we describe the construction of six independent NotI linking l
ibraries specific for the total human genome. These libraries were mad
e using three different vectors and two combinations of restriction en
zymes. Altogether, these six libraries contain more than 1 million rec
ombinant phages. Considering that the human genome contains about 3000
-5000 NotI sites, it is likely that all clonable NotI sites are presen
t in these libraries. Two of the six libraries were transferred into p
lasmid form. At the same time, a chromosome 3-specific EcoRI-NotI libr
ary (NRL1) was constructed. This library considerably increases the re
presentation of cloned NotI sites in combination with previously const
ructed libraries that were made using BamHI-NotI digestion. All librar
