A quick and sensitive dot-blot assay using non-radioactive labelled RN
A probes was developed for the detection of the CF53 strain of hepatit
is A virus (HAV) in cell culture. The cDNA of the 5' end of the HM175
strain was inserted in a transcription vector pSPT18 and was used to s
ynthesize P-32- or digoxigenin-labelled RNA probes. These RNA probes s
pecifically detected the RNA of the CF53 strain and can be used to det
ect HAV in PLC/PRF/5 cells. The sensitivity of non-radioactive tests w
as comparable to that of radiolabelled probes.
