We report a new synthetic strategy for the preparation of the amyloido
genic protein fragment beta A4, through an easily purified (and charac
terized) intermediate of high aqueous solubility. This novel intermedi
ate consists of beta A4(1-43) with backbone amide protection, provided
by the N-(2-hydroxy-4-methoxybenzyl) group, on residues Phe(20) and G
ly(25,29,33,38). The new strategy enabled derivatized beta A4(1-43) to
be isolated and purified by standard reverse-phase HPLC procedures an
d characterized by electrospray mass spectrometry (ESMS).
