Tordon herbicide (4-amino-3,5,6-trichloropicolinic acid -picloram, and
2,4-dichlorophenoxyacetic acid - 2,4-D) depresses the phosphorylation
efficiency of rat renal cortical mitochondria as deduced from the dec
rease of respiratory control (RCR) and ADP/0 ratios when NAD+-dependen
t substrates were used; NADH oxidase (50%) and NADH cytochrome c reduc
tase (23%) were also inhibited. Tordon had no effect on the other enzy
matic complexes of the respiratory chain. The amplitude and the veloci
ty of swelling induced by glutamate is progressively inhibited, while
with succinate, TMPD (N,N,N1,N1-tetramethyl-p-phenylediamine) + sodium
ascorbate and ATP as substrate a partial inhibition of the amplitude
was observed. Tordon promote a decrease in the membrane potential upon
energization of the mitochondrial membrane only by glutamate. These r
esults characterize an interaction of Tordon with complex 1 of proton-
motive force of the mitochondrial inner membrane without affecting its
elasticity.
