Codon recognition by Escherichia coli tRNA(4)(Leu) and tRNA(5)(Leu) wa
s investigated by analysis of the competition between two aminoacyl-tR
NA species in an in vitro protein synthesis. Both tRNA species strictl
y obey the wobble rule when they are in competition with other tRNA sp
ecies. This is probably due to the post-transcriptional modifications
at the first position of the anticodon of these tRNA(Leu) species, sup
porting the proposal that the conformational rigidity of post-transcri
ptionally modified pyrimidine nucleotides guarantees the correct codon
recognition.
