The gene of human angiotensin II type 2 (AT(2)) receptor was isolated
from a genomic DNA library prepared from human placenta. The coding re
gion of the human AT(2) receptor gene was contained in a single exon c
oding segment of the gene indicating an intronless structure of the co
ding region. The amino acid sequence of human AT(2) receptor deduced f
rom its nucleotide sequence has 363 amino acids and shows a high degre
e of sequence identity to rat and mouse receptor sequences. Specific b
inding of [I-125]Sar(1)Ile(8)-angiotensin II was demonstrated in COS-7
cells transfected with a plasmid containing the human AT(2) sequence.
Scatchard analysis and ligand displacement profile were typical of th
e AT(2) receptor. Reverse transcription-polymerase chain reaction anal
ysis showed that AT(2) receptor mRNA was expressed in adult uterus and
