Acetobacter polyoxogenes was transformed with plasmid DNA by electropo
ration. The following points were essential for transformation: (i) di
lution of the culture broth with cold water and air bubbling of the cu
lture broth for transformation at discharging from a jar fermenter, an
d (ii) selection of transformants by liquid cultivation. For shortenin
g of the lag time in cultivation for selection of transformants, the f
ollowing treatments were useful: (i) addition of sucrose to the cell s
uspension during transformation and to the broth for cultivation, and
(ii) addition of 1 mM MgCl2 to a mixture of cells and DNA during elect
roporation.
