Abstract
Improved conditions for non-radioisotopic DNA fingerprinting analysis using digoxigenin M43 probe were developed. Highly resolved banding pa tterns were obtained with specific-primed labelling of M43 DNA and Pho togene membrane as a blotting medium under low stringency of hybridiza tion conditions and post-hybridization washes. DNA fingerprinting of o ne family with 4 children and a pair of identical twins were performed and the signals were detected by both colourimetric and chemiluminesc ent detection systems. Readable pattern was generated with as little a s 0.5 mug of human genomic DNA This non-radioisotopic DNA fingerprinti ng was simple, reliable and should be applicable as an alternative to the isotopic method.
