By using immunogold electron microscopy technique we have demonstrated
the presence of GABA and glutamate in the microvesicles of rat neuroh
ypophysis. The distribution of neurotransmitters was revealed using mo
noclonal antibody against glutamate and polyclonal antibody against GA
BA with colloidal gold labelling. Distribution of gold particles was s
imilar for both neurotransmitters being maximal over heterogenous popu
lations of microvesicles and mitochondria. After 10 min cerebral ische
mia immunolabelling of microvesicles was more intense, especially for
glutamate. The role of glutamate toxicity after ischemia is discussed.
