The HindII-HindII fragment of probe pH2-42 of human genome locus D13S2
5 was sequenced, and its localization in the q14-q21 region of chromos
ome 13 confirmed by in situ hybridization. Seven oligonucleotide prime
rs for PCR were selected to amplify fragments almost completely coveri
ng the pH2-42 sequence. The data of Bowcock and Hebert [Nucl. Acids Re
s., 18, 7194 (1990)] and of the present work were used to reconstruct
the distribution of polymorphic SspI sites in D13S25. This makes the p
rimer set applicable not only for screening cosmid and YAC libraries b
ut also for marking the given locus of human chromosome 13.
