The interleukin-2 receptor beta-chain (IL-2Rbeta) is responsible for t
he IL-2 internalization and signal transduction. To identify domains f
rom the IL-2Rbeta extracellular region which are involved in ligand-re
ceptor interactions, eight peptides of 8 to 25 amino acids correspondi
ng to IL-2Rbeta sequence 30-54 were synthesized. Their biological effe
ct was tested with [I-125]IL-2 equilibrium binding to PHA-blasts. Prei
ncubation of [I-125]IL-2 with peptides IL-2Rbeta (35-54) and (30-42) a
ffects IL-2 binding to both high and intermediate affinity receptors b
y revealing a greater number of active sites with lower affinity.
