The action of platinum anticancer agents (cisplatin, oxoplatin and cyc
loplatam) on the physicochemical characteristics of artificial and pla
sma membranes was studied. The drugs in question changed the electric
resistance of artificial phospholipid membranes, electric potential an
d surface tension of the water - phospholipid boundary. Drugs action g
rew with their concentrations; cisplatin proved the most effective. In
vivo injected cisplatin action on the hepatocytes plasma membranes pr
operties was studied by binding the fluorescent probe - magnesium salt
of 8-anilinonaphthalene sulfonic acid (ANS). Binding constant and bin
ding sites concentration changes were observed 15 min, 1 hour, 4, 24 a
nd 96 hours after the drug injection, a character of the changes being
different for the intact rats and the rats with the Guerin carcinoma.
The conclusion is drawn that platinum compounds display a surface act
ivity and that their biological effects are highly connected with thei
r action on the membrane structural and functional state.
