To elicit antibodies directed specifically against the neuron-specific
form of the c-sre gene product, pp(60c-src(+)), we used an antigenize
d antibody comprising a decamer containing the amino acid sequence spe
cific to pp(60c-src(+)) inserted into the third hypervariable loop of
the heavy (H)-chain variable (V)-region. This was used to raise anti-i
diotype antibodies reacting with the peptide epitope in rabbits. The a
ntisera reacted with pp(60c-src(+)), as judged by immune blotting, imm
unoprecipitation, immune complex kinase assay, and indirect immunofluo
rescence staining, but did not react with the fibroblast form of the c
-sre gene product, pp(60c-scr). Antigenized antibody is a useful appro
ach for producing antibodies able to distinguish between isoforms of t
he same gene product and specific for the neuronal form of the Src pro
tein.
