Transgenic peanut plants were produced using Agrobacterium mediated ge
ne transfer. Primary leaf explants of peanut were co-cultivated with A
grobacterium tumefaciens LBA 4404 harbouring the binary plasmid pBI 12
1 (conferring beta-glucuronidase activity and resistance to kanamycin)
and cultured on regeneration medium supplemented with kanamycin to se
lect putatively transformed shoots. They were rooted and plants were t
ransferred to soil. Stable integration and expression of the transgene
s were confirmed by NPT II assay, Southern blot hybridization and GUS
assay.
