Vital fluorochrome hydroethydine was used for measuring relative extra
- and intracellular production of active oxygen in phagocyting blood c
ells in hemoglobinopathies. Hydroethydine sensitivity to diverse free
oxygen forms was studied in various model systems. Probability of fals
e-positive results was estimated. It was established that hydroethydin
e interacts with superoxide anion radicals in molar ratio 1:1, that in
tracellular production of active oxygen forms in leukocytic mass cells
stimulated by phorbolmyristatacetate was enhanced in patients with ma
jor beta+-thalassemia and in a patient with hemoglobinopathy due to un
stable hemoglobin Hb-Alesha. In thalassemia minor no significant diffe
rences compared to control were reported. Potential causes and value o
f the results obtained are under discussion.
