A method for the determination of L-697, 661, a human immunodeficiency
virus type 1 reverse transcriptase inhibitor, and its 6-hydroxy metab
olite in plasma is described. Following analyte isolation via solid ph
ase extraction, samples are analyzed by HPLC using a column switching
system. Column switching allows each analyte to be simultaneously anal
yzed under isocratic reverse phase conditions in a run time of 21 minu
tes. The assay was linear in the concentration range of 10-500 ng/mL f
or each analyte when 1 mL aliquots of plasma were extracted. The assay
has been fully validated and utilized for human pharmacokinetic studi
es.
