The carbohydrate structures of blotted glycoproteins can be analyzed b
y probing with lectins. The objective of the present work was to optim
ize the lectin blotting of human plasma glycoproteins separated by two
-dimensional gel electrophoresis and the detection by the sensitive ch
emiluminescence method. The proposed detection method was found to be
ten times more sensitive than a standard colorimetric reaction. Furthe
rmore, the generated signals are detected on a X-ray film and provide
a permanent record. The method is also very reliable when compared to
the colorimetric detection. The present procedure for glycoprotein ana
lysis is particularly well suited for screening changes in glycosylati
