Abstract
The carbohydrate structures of blotted glycoproteins can be analyzed b y probing with lectins. The objective of the present work was to optim ize the lectin blotting of human plasma glycoproteins separated by two -dimensional gel electrophoresis and the detection by the sensitive ch emiluminescence method. The proposed detection method was found to be ten times more sensitive than a standard colorimetric reaction. Furthe rmore, the generated signals are detected on a X-ray film and provide a permanent record. The method is also very reliable when compared to the colorimetric detection. The present procedure for glycoprotein ana lysis is particularly well suited for screening changes in glycosylati on of proteins in biological samples. (C) 1994 Academic Press,Inc.
