B. Venkaiah et A. Kumar, EGG-SHELL BOUND STARCH PHOSPHORYLASE PACKED-BED REACTOR FOR THE CONTINUOUS PRODUCTION OF GLUCOSE 1-PHOSPHATE, Journal of biotechnology, 36(1), 1994, pp. 11-17
A process for the continuous production of glucose 1-phosphate, a cyto
static compound used in the therapy of cardiopathy, from starch, is de
veloped. Starch phosphorylase from sorghum leaves has been immobilized
on various solid supports, viz. polyacrylamide, gelatin, egg shell an
d alumina. For the continuous production of glucose 1-phosphate, egg s
hell bound enzyme is found suitable over other matrices bound enzyme.
Egg shell bound enzyme showed maximum thermostability (half-life, 14 h
) at 40 degrees C and it can be reused many times for the production o
f glucose 1-phosphate. Product formed was purified by adsorption and i
on exchange chromatography using Norite and DEAE cellulose columns.
