Basal membrane turnover implies the regulation of specific metalloprot
eases (MMP) with cleavage abilities for collagen IV. We have set up a
model of neutrophil (PMN)-endothelial interaction in vitro which allow
ed to observe a net gelatinase exocytosis when PMN. adhered to plastic
substrates. When PMN adhered to endothelial monolayers, this activity
was significantly diminished (-30%; p<0.05) and furthermore, RNA tran
scripts for MMP were specifically down-regulated. Our study points to
an endothelial inhibitory regulation of matrix remodeling during PMN-e
ndothelial interaction.
