For more precise location of the gene for palmoplantaris hyperkeratosi
s (PPHK) on the large arm of chromosome 17, a multilocus mapping was p
erformed using eleven highly polymorphic microsatellite and VNTR loci
overlapping the 17th chromosome. The most probable location of the PPH
K gene is between the loci D17S800 and D17S791. However, the adjacent
telomere and centromere intervals, D17S791 - D17S806 and D17S800 - THR
A1, could not be excluded as possible regions of the PPHK gene. The ti
ght linkage of the PPHK gene with the cluster of keratin genes in 17q
(z1 = 8.36 at THETA = 0.00) was confirmed by a study of linkage of the
PPHK gene with the KRT10 locus (insertion-deletion polymorphism in th
e 7th exon). No mutations in the first exon of the keratin 10 gene wer
e revealed by sequencing of this exon and its flanking intron sequence
