A rapid HPLC method for determining protoporphyrin is employed in the
procedure described for measuring coproporphyrinogen III oxidase (CO)
activity in bacteria. The activity was found to be linear with protein
up to 300 mug/assay and with incubation time up to 2 h. CO activities
measured in Escherichia coli, Pseudomonas aeruginosa and Klebsiella p
neumoniae were found to be 237, 128 and 22 pmol protoporphyrin/mg prot
ein/h, respectively.
