Mycobacterium leprae was isolated from skin-punch biopsies of 2 untrea
ted lepromatous leprosy patients. The bacteria were enumerated, dilute
d 10-fold and cultured in Middlebrook 7H9 medium supplemented with alb
umin, dextrose, catalase and C-14-palmitic acid. The cultures were inc
ubated at 33-degrees-C in a modified Buddemeyer radiorespiratory detec
tion vessel. Those cultures containing at least 10(7) mycobacteria dem
onstrated a progressive evolution of (CO2)-C-14.
