Monoclonal antibodies (mAbs) raised against heavy and light chains of
goldfish immunoglobulin (Ig) were characterized by a Western blot tech
nique. A complete cross-reactivity was observed between carp and goldf
ish Ig. These mAbs were used for the quantification of carp Ig and ant
i-Yersinia ruckeri antibodies by ELISA. An ELISPOT assay was also deve
loped in carp to quantify Ig-secreting cells (ISC) and antibody-secret
ing cells (ASC). The number of ASC was maximum on day 18 post-vaccinat
ion and decreased to the basal level on day 28. The antibody levels in
sera were maximum on day 18 and slowly decreased until day 28.
