For quantification of histamine, we developed a highly specific and se
nsitive method of postcolumn fluorescence detection HPLC using a tetra
hydrofuran solution of orthophthalaldehyde. Its detection limit was 2
- 5 pg. This method was applied to the measurement of histamine in hum
an blood and urine samples. Analysis by LC/MS and NMR suggested that t
he fluorophore in this method has an isoindole structure.
