Some modifications of HPLC method for identification 1-hydroxypyrene i
n rat urine are described. As opposed to analytical procedure presente
d in literature which includes solid phase extraction (SPE) and HPLC d
etermination with fluorescence detection, more approachable extraction
with cyclohexane and detection in the UV range were used after enzymi
c hydrolysis. 1-Hydroxypyrene could be detected in the urine of rats w
ith 88.3 % average recovery level and 0.25 mug l-1 urine detection lim
it. After pretreatment rats with pyrene p.o. in dose 20 mg kg-1 excret
ion of 1-hydroxyprene during 72 h increased in urine from 0.22 to 105.
7 mug per mg of creatinine.
