Evidence is reported here that alkalinization of Clostridium acetobuty
licum cytoplasm involves hydrogenase activity. A decrease of in vivo h
ydrogenase activity is accompanied by intracellular accumulation of pr
otons leading to a negative (interior acidic) pH gradient. How ever, t
he organism is able to maintain a constant proton motive force by inte
rconverting chemical and electrical potentials.
