Abstract
High cell density culture is essential for the efficient production of biological substances such as monoclonal antibody and hormones by cul tured animal cells, since animal cells usually have a low growth rate and produce small amount of product. Novel cell immobilization methods using photo-crosslinkable resins and macroporous cellulosic supports were developed to achieve high cell density. The hybridoma and recombi nant L-929 cells immobilized were cultured in bioreactors and the prod uctivities of monoclonal antibody and erythropoietin were improved ver y much. In hybridoma cell culture using serum-free medium, the effects of dissolved oxygen concentration and growth substrate concentrations such as glucose and glutamine were examined with respect to the metab olism and antibody production. Furthermore, the separation and concent ration method of monoclonal antibodies by aqueous two-phase extraction using Pluronic F68 was proposed.
