The purpose of this investigation was to improve the solubility of spi
ronolactone (1) for use in oral pediatric formulations. The cosolvent
approach was used to solubilize 1, using combinations of ethanol, prop
ylene glycol, polyethylene glycol 400, glycerin, and 70% w/w sorbitol.
The solubility of 1 in several cosolvent blends was determined. The r
esults obtained indicate that the solubility of 1 can indeed be increa
sed by as much as 70-fold, by the cosolvent approach. Certain pairs of
solvents exhibited a synergistic increase in solubility. The Yalkowsk
y equation was used to model the solubility increases. Correlations be
tween solubility and the dielectric constant of the cosolvent blends w
ere investigated.
