Abstract
The nucleotide sequence of E1A and E1B region of simian adenovirus SA7 (C8) integrated in malignant SH2 rat cell Line was studied. At least t wo copies of the SA7 oncogene integrated in the SH2 genome were reveal ed by Southern blot hybridization. Polymerase chain reaction analysis of E1A and E1B regions showed heteroduplex structures, proving differe nces between the integrated copies. The heteroduplex molecules of diff erent electrophoretic mobility were separated, and the nucleotide sequ ence of the chains corresponding to different copies was determined by the modified method of Sanger. The copies differed mainly in the micr osatellite regions: EIA positions 894-902, (GCG)3/(GCG)4, and E1B posi tions 2037-2048, (GCA)3/(GCA)4. All the deviations found pertain to th e coding regions of the SA7 oncogene.
