Abstract
A full-length cDNA copy (PLRV(fl)) of potato leafroll virus (PLRV) was constructed and examined in vivo for its biological activities by tra nsient expression experiments with plasmid DNA or in vitro transcribed RNA. In addition, PLRV(fl) cDNA was stably introduced into the genome of potato plants by Agrobacterium-mediated leaf disc transformation. Both transient and stable expression of PLRV(fl) resulted in the synth esis of genomic and subgenomic PLRV RNAs. Transgenic plants accumulate d the 17-kDa movement protein and displayed the typical symptoms of PL RV infection. This is the first example of the constitutive expression of a phloem-limited virus in planta.
