A rapid and simple method for determination of fluconazole in plasma s
amples by high performance liquid chromatography was developed. The me
thod includes a single extraction of alkalinized plasma with ethyl ace
tate. Plasma extracts were analyzed on a reverse-phase column eluted w
ith a mixture of acetonitrile and 0.05 M sodium monohydrogen phosphate
(pH 4) and detected by absorbance at 210 nm. Retention times for fluc
onazole and the internal standard were 7 and 13 min respectively. The
method was linear in the range of 0.1 to 5 mu g/ml and the detection l
imit of the method was 20 ng/ml. This method is suitable for determina
tion of fluconazole after administration of the drug at therapeutic do
ses, and it could be used in pharmacokinetic studies of the drug in hu
mans.
