Abstract
AIM: To evaluate anti-hepatitis drugs. METHODS: Mice were injected iv with viable BCG 5x10(7) (live) bacilli/mouse, after 10 d, iv lipopolys accharides (LPS) 5-10 mu g/mouse. Serum aminotransferase levels and li ver tissue were examined 10 h after iv LPS. Sixteen drugs were evaluat ed in this model. RESULTS: The level of AlaAT and AspAT were increased markedly. Submassive necrosis and infiltrations of granulocytes and l ymphocytes were seen, which were not recovered till 14 d after iv LPS. Administration of interferon alpha-2a, Ara-AMP, hepatocyte growth fac tor, biphenyl dimethyl dicarboxylate, bicyclol, prednisolone, and cycl ophosphamide for 10 d prior to iv LPS suppressed the elevation of seru m AlaAT and AspAT. CONCLUSION: The model of immunological liver injury reproduced by iv BCG + LPS in mice may be used for evaluating antihep atitis drugs.
